Title of Method

Quantitation of Six Common Food Sugars and Inositol by HPLC-MS

AOAC Candidate Method Number (e.g. ALN-01)

SUG-004

Applicable SMPR

2018.001

I. Summary of the Method

The method quantifies six common food sugars and myo-inositol. Extraction is accomplished with 25% reagent alcohol in water at 60 C. Protein is precipitated using Carrez procedures, followed by centrifugation and dilution. The analysis is completed using LC-MS. The method is applicable to the sugar analytes listed in the SMPR. The submission does not address potential interferences as outlined in the Applicability section of the SMPR. Specifically isobaric interferences should be addressed in this method.

II. Review of the Method Only 1. Does the applicability of the method support the applicability of the SMPR? If not, please explain what is missing. 2. Does the analytical technique(s) used in the method meet the SMPR? If not, please specify how it differs from what is stated in the SMPR. 3. Are the definitions specified in the SMPR used and applied appropriately in the method? If no, please indicate how the terms are used.

The analytical technique is appropriate to meet the SMPR.

Is a pH adjustment step needed during the Carrez procedure?

Relatively low sample weight (0.5 g) may impact precision for heterogeneous matrices.

I did not see a representative chromatogram in the submission.

Although the exact same matrix terms are not used, the method conveys the intent of the SMPR defined matrix categories. Yes, the method applies the SMPR definitions appropriately.
