Biotyper (MBT). The Id-Fungi Plates contain a membrane filter to facilitate mold harvesting. The workflow combining this new plate format with the MBT was evaluated by two laboratories on 125 strains. A software module has been developed to include adapted thresholds for identification of molds; 856 mass spectra were generated from 56 strains from 37 species and 18 genera. The MBT Reference Library for fungi identification have been extended using strains from 26 laboratories and 10 countries in order to cover a relevant biodiversity. The Id-Fungi Plates enable to collect the biomass in less than 10 seconds for almost 100% of the tested strains, while more than 1 minute is usually required for 26% of the strains cultivated on Sabouraud agar. 98.7% of successful acquisition rates were acquired with the new Software Module. The Library 3.0 covers 577 reference spectra. The Id-Fungi plates improves the filamentous fungi sampling with the MALDI Biotyper. The Filamentous Fungi Module optimizes data acquisition and analysis, improving mass spectrum acquisition up to 98.7%. The Library 3.0 has been expended to 180 species from 62 different genera. Validation of the foodproof ® Yeast & Mold Quantification LyoKit showed results in <5 hours equivalent to or better than ISO 6611 for the detection and quantification of yeasts and molds in dairy products. Dilution series of genomic DNA from 10 yeast and mold species were tested with 3 replicates. Even with 1 genome equivalent of DNA were all samples positive. For inclusivity 236 species were positively tested and more than 50 bacteria, plants and mammal cells were tested negative. The kit specificity is 100%. The foodproof ® Yeast and Mold Quantification LyoKit detects and quantifies yeast and molds in one single test in shortest time. In combination with the food- proof ® StarPrep Two Kit validated protocols for all kinds of dairy products from finished products to raw materials are available. In all protocols a live/dead differentiation of cells with Reagent D is included. The LoD (95%) was determined at 80 cfu/g sample. For high-throughput analysis the newly developed foodproof ® StarPrep Six 8-Strip Kit can be used for DNA isolation. This enables fast and easy DNA extraction from yeasts and molds using 8-strips and a multichannel pipette. With a low hands-on time up to 96 samples can be processed in parallel. Additionally new protocols for other sample matrices like cannabis, soy or chili sauce and seafood are now available.
