AOAC Official Method 2018.02 Enumeration of Yeast and Mold in Select Foods MC-Media Pad™ Yeast and Mold Device First Action 2018 (Applicable to the enumeration of yeast and mold from breaded chicken nuggets, dry pet food, orange

juice concentrate, yogurt and cake mix).

A. Principle The MC-Media Pad YM is intended to enumerate viable yeast and mold in select foods in as little as 48

hours. The MC-Media Pad YM uses a special medium composition and unique redox indicator for

detection. Once the liquid sample is inoculated onto the test pad, the sample diffuses through the whole

pad by capillary action. The medium reconstitutes automatically. If target organisms are present, they

grow red colored colonies on the test pad.

B. Apparatus and Reagents a. MC - Media Pad Yeast & Mold – 100 test pads (25 pads/pouch, 4 pouches/box) Available from MilliporeSigma #1323030001. b. Sterile Diluent- 0.1% Peptone water & Phosphate Buffered Saline c. Pipettes- capable of pipetting 1,000 µL or a serological pipette d. Sterile pipette tips- capable of 1,000 µL e. Laboratory paddle-blender- Seward 400 or equivalent f. Filter Stomacher bags - Seward or equivalent g. Incubators – Capable of maintaining 25 ± 1 o C h. Refrigerator - capable of maintaining 2-15 o C, for storing the MC-Media Pads
