Sesame ELISA validation approach

Three commercial sesame ELISA kits were selected. Selected based on previous use at FDA, and availability of internal validation study reports. ELISA kits were run as per the kit instructions. Sesame incurred/spiked model foods were prepared. Cookies, hummus, and spice mix Hummus was analyzed on day 1 and day 7 of storage at 4C. Incursion/spike levels- 0, 5, 10 and 30 ppm sesame. Ground white sesame seed was used as the reference material for spiking/incursion. Triplicate test potions per matrix were analyzed by the three ELISAs. Three analysts performed the evaluation.
