16. CONCLUSION

The results of these studies demonstrate that an eight-cycle increase in the processing

time of the BAX® System Real-Time PCR Assay for Salmonella does not affect the

AOAC INTERNATIONAL previously certified method, except to improve the sensitivity for low-level positive samples around the limit of detection. This was demonstrated by showing no change in the results for a variety of matrices spiked at a fractional level, through inclusivity and exclusivity, and

through documentation of fluorescent probe performance during the additional cycles for

samples that contain and those that do not contain Salmonella . It is recommended that the

modification to PTM #081201 be approved.
