Transfer each sample lysate into individual Reagent tubes first followed by the NC. Hydrate 1 the RC tube last. 2 5. Use the 3M™ Molecular Detection Cap/Decap Tool-Reagent to decap the Reagent tubes – 3 one Reagent tube strip at a time. Discard cap. 4 5.1 Transfer 20 µL of Sample lysate from the upper ½ of the liquid (avoid precipitate) in the 5 LS tube into corresponding Reagent tube. Dispense at an angle to avoid disturbing the 6 pellets. Mix by gently pipetting up and down 5 times. 7 5.2 Repeat step 5.1 until individual Sample lysate has been added to a corresponding 8 Reagent tube in the strip. 9 5.3 Cover the Reagent tubes with the provided extra caps and use the rounded side of the 10 3M Molecular Detection Cap/Decap Tool-Reagent to apply pressure in a back and 11 forth motion ensuring that the cap is tightly applied. 12 5.4 Repeat step 5.1 as needed, for the number of samples to be tested. 13 5.5 When all sample lysates have been transferred, repeat 4.1 to transfer 20 µL of NC 14 lysate into a Reagent tube. 15 5.6 Transfer 20 µL of NC lysate into a RC tube. Dispense at an angle to avoid disturbing 16 the pellets. Mix by gently pipetting up and down 5 times. 17 6. Load capped tubes into a clean and decontaminated 3M Molecular Detection Speed Loader 18 Tray. Close and latch the 3M Molecular Detection Speed Loader Tray lid. 21 22 7. Review and confirm the configured run in the 3M Molecular Detection Software. 23 8. Click the Start button in the software and select instrument for use. The selected 24 instrument’s lid automatically opens. 25 9. Place the 3M Molecular Detection Speed Loader Tray into the 3M Molecular Detection 26 Instrument and close the lid to start the assay. Results are provided within 75 minutes, 27 although positives may be detected sooner. 28 AOAC Research Ins itute 19 20
