Assay design: considerations • Intended use ‐ Diagnostic (broadly inclusive) vs quantitative (amplification efficiency)  applications ‐ How specific does the assay need to be? • Ex. Brucella spp.  is very monophyletic   • Sequence availability ‐ Limited or extensive information? • Design considerations based reagents used ‐ Mastermix; the type of probe used, etc.
