AOAC SPIFAN Nutrients ERP Review (March 14, 2019)

2016.13 (Jan. 2019) Carot-02 MLT Report FOR ERP USE ONLY DO NOT DISTRIBUTE

electronic template provided by the study director. Ten of the participating laboratories submitted data before the deadline. Data Review and Reporting MLT participants were provided with an electronic template (Excel spreadsheet) for reporting data, including the LC system used, standards used, weights, and LC peak areas. Peak retention times and peak widths from system suitability standards were used to determine resolution. The reporting template was designed to perform all calculations, including linearity and sample carotenoid concentrations, specified in the analytical method. A response factor of 1.4 instead of 1.7 was used for 13-cis-lutein and 13ʹ-cis-lutein, based on feedback from ISO review and further testing. Carotenoid concentrations that were originally reported as µg/100 g on a powder basis were converted to a reconstituted basis using the factor of 25 g powder/225 g reconstituted product. After the data was compiled, it was coded according to laboratory. Standard response factors and linearity were reported for each analyte. Carotenoid concentrations for each matrix were plotted and raw data was analyzed with the AOAC International Interlaboratory Study Workbook for Blind Replicates, version 2.1. Outliers were identified using the Grubbs and Cochran tests. Repeatability (RSD r ), reproducibility (RSD R ), and HorRat values were calculated and compared with SMPR limits and expected values. Acknowledgments The study director would like to acknowledge the following collaborators for their efforts in setting up the method and providing the data for the completion of this study. Their participation and feedback on the method is greatly appreciated.
