I. Summary of the Method

The method abstract says that the method is to quantify A-type PACs in cranberry products. The first step of the method isolates the total PAC fraction by loading an aqueous extract on a sephadex LH-20 column, eluted to remove the anthocyanins (as monitored by absorbance at 520 nm) then the PAC are eluted with acetone:water (7:3). Samples and calibrants were then injected on a diol column, eluted with a gradient of acetonitrile:acetic acid (98:2) and methanol:water:acetic acid (95:3:2). Fluorescence detection was used with an excitation wavelength at 230 nm and emission at 321 nm.
