Preparation First Action to Final Action at AOAC -------------------------------------------------------------------------------------------------------------------------------------- Method AOAC OMA #2017.09 Confirmation and Identification of Salmonella spp., Cronobacter spp., Campylobacter spp. and Other Gram-Negative Organisms by the Bruker MALDI Biotyper Method First Action 2017.09 Method AOAC OMA #2017.10 Confirmation and Identification of Listeria monocytogenes , Listeria spp., and Other Gram-Positive Organisms by the Bruker MALDI Biotyper Method First Action 2017.10 Introduction The Bruker MALDI Biotyper (MBT) method is designed to rapidly confirm (meaning to confirm the presumptive result of an alternative or a reference method) and identify (meaning to determine the identity of an analyte from a variety of agar plates). The method was validated for use with common nonselective, selective, and chromogenic agars following isolation of the organisms per the appropriate reference or proprietary method. Samples are analyzed using matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) MS. Two hardware instruments can be used in the MBT configuration: the microflex LT/SH and the microflex LT/SH smart. The highly abundant microbial ribosomal proteins result in a mass spectrum with a characteristic mass and intensity distribution pattern. The generated spectrum pattern of these proteins is used to reliably and accurately identify a particular microorganism by matching against a library (version MSP 6903 or higher). The library is updated every year. The MALDI Biotyper was first validated in December 2017 with two different Official Methods of Analysis (OMA): - Method AOAC OMA #2017.09: Confirmation and Identification of Salmonella spp., Cronobacter spp., and Other Gram-Negative Organisms by the Bruker MALDI Biotyper Method: Collaborative Study - First Action 2017.09 - Method AOAC OMA #2017.10: Confirmation and Identification of Listeria spp., L. monocytogenes and Other Gram-Positive Organisms by the Bruker MALDI Biotyper Method - First Action 2017.09 The scope of the Method AOAC OMA #2017.09 was extended to confirmation of Campylobacter spp. in June 2017, in order to get the following recognition:
