SUMMARY OF COMMENTS  OMAMAN‐46: Enumeration of E. coli and Coliform

ER 5 It is an economic method that does not need specific instruments for its performing, so it is very  accessible to laboratories of low complexity ER 6 Manuscript is of normal quality ER 7 Generally well written and clearly presented. ER 8 Well written. ER 9 Well written and thorough Cons/Weaknesses of the Manuscript: ER 1 N/A ER 2 On page two of the collaborative manuscript, line 2.  Is it necessary to individually list the  incubation temperatures?  Practically, the plates can be incubated at 30‐37C +/‐ 1C. ER 3 Wondering if a highly contaminated product of some pathogens lines 1‐3 as described on page 7  may cause any issues. ER 4 none ER 5 No ER 6 NA ER 7 The title implies that an E. coli count and a total coliform count can be obtained. This does not  seem to be exactly so. A total coliform count can be obtained (blue plus red+/‐gas colonies).  However, the blue+/‐gas only count will miss glucuronidase negative E. coli O157 but will include  atypical non‐E. coli coliforms (glucuronidase positives). Suggest changing title to "Evaluation of 3M  Petrifilm Rapid E.coli/Coliform Count Plate for enumeration of total coliforms and enumeration of  glucuronidase positive E. coli plus atypical non‐E. coli coliforms: Collaborative Study." Also could  pick 10 blue colonies and check if any are atypical non‐E. coli coliforms.
