Protocol with an enrichment step at +41.5°C

◦ 25 g of sample. ◦ 225 mL of buffered peptone water (BPW). ◦ Mix using a paddle blender or manually for environmental surface samples. ◦ Incubate at +41.5°C ± 1°C for 18 ‑ 26 hours. Note: For environmental surface samples, add enough BPW to cover the sampling tool (for example 10 mL for a swab, 100 mL for a sponge, 225 mL for cloth). ◦ 25 g of sample. ◦ 225 mL of preheated (+41.5 ± 1°C) buffered peptone water (BPW). ◦ Mix using a paddle blender. ◦ Incubate at +41.5°C ± 1°C for 8 ‑ 24 hours. Protocol for 50–375 g ◦ X g of sample. ◦ 3X mL of preheated (+41.5°C ± 1°C) buffered peptone water (BPW). ◦ Mix using a paddle blender. ◦ Incubate at +41.5°C ± 1°C for 10 ‑ 24 hours. ◦ 25 g of sample. ◦ 225 mL of buffered peptone water (BPW) or specific enrichment broth according to ISO 6579 and 6887. ◦ Mix using a paddle blender for 1 minute or manually for swabs and sponges. ◦ Incubate at +37°C ± 1°C for 18 ‑ 26 hours. Note: For environmental surface samples, add enough BPW to cover the sampling tool (for example 10 mL for a swab, 100 mL for a sponge, 225 mL for cloth). Protocol for 50–375 g ◦ X g of sample. ◦ 9X mL of preheated (+37°C) buffered peptone water (BPW). ◦ Mix using a paddle blender. ◦ Incubate at +37°C ± 1°C for 20 ‑ 28 hours. ◦ Dilute 1/10 in peptone salt or physiological saline before testing. Protocol for 50–375 g ◦ X g of sample. ◦ 9X mL of preheated (+37°C) enrichment broth according to ISO 6579 and ISO 6887. ◦ Mix using a paddle blender. ◦ Incubate at +37°C ± 1°C for 22 ‑ 30 hours. ◦ Dilute 1/10 in peptone salt or physiological saline before testing. Protocol for 50–375 g ◦ X g of sample. ◦ 3X mL of buffered peptone water (BPW) + novobiocin (10 mg/L). ◦ Mix using a paddle blender. ◦ Incubate at +37°C ± 1°C for 20 ‑ 28 hours.
