Standard procedure for a 25 g sample for a variety of foods, including fresh raw ground chicken, fresh raw chicken breast, fresh raw fish, creamy peanut butter, vanilla ice cream, and dry pet food
◦ 25 g of sample. ◦ 225 mL of buffered peptone water (BPW) at room temperature (BPW or mTSB for ground chicken). ◦ Homogenize and incubate at +42°C ± 1°C for 18 ‑ 24 hours. ◦ 30 mL of sample. ◦ 30 mL of preheated buffered peptone water (BPW). ◦ Homogenize and incubate at +42°C ± 1°C for 18 ‑ 24 hours. ◦ 375 g of sample. ◦ 1125 mL of preheated buffered peptone water (BPW) or mTSB (only BPW for ground bison). ◦ Homogenize and incubate at +42°C ± 1°C for 10-24 hours. ◦ 375 g of sample. ◦ 1125 mL of preheated buffered peptone water (BPW). ◦ Homogenize and incubate at +42°C ± 1°C for 22 ‑ 26 hours. ◦ 5 enriched samples can also be wet ‑ pooled for analysis (see Standard Procedure 2). Standard Procedure 2 : After incubation, manually mix the contents of the blender bag. Optionally, a sterile technique can be used to remove 1 mL of enriched sample and place it in a pre ‑ labeled microcentrifuge tube. Note: For the wet pooling protocol 375 g Romaine lettuce, combine 1 mL from 5 test sample enrichments in a pre ‑ labeled tube.
