OMAMAN-20 C/ In House Validation Report ERP Use Only - March 2015

Since only a prolamin extract was tested as a single determination, three other lots were checked with matrix-containing samples (table 8 and 9) and 20 replicates per concentration. Results for ethanol extraction in table 8 reveal that all three lots are comparable. There is only a very small variation for the POD values at 1 mg gliadin per kg and ethanol extraction. Results are identical for 0.0, 0.5 and 2.0 mg/kg.

Table 8 . Lot-to-Lot comparability for rice flour spiked with WGPAT gliadin and extracted with 60% ethanol; results for 20 replicates per concentration level are given.

Comparable results were obtained when a processed cookie sample after Cocktail extraction was used (table 9). Due to the higher dilution of the extracted sample before measurement, the concentration which indicates the extent of variation is at 2 mg/kg compared to 1 mg/kg for the sample extracted with ethanol. Again, all three lots are highly comparable.
