Determination of concentration levels

The concentration level testing for all incurred or contaminated samples (4.1.6.2) will be performed with the reference ELISA test kit RIDASCREEN® Gliadin R7001 (AOAC OMA first action 2012.01). The minimum possible concentration will thus be 2.5 mg gliadin/kg sample, since this is the limit of quantification of this method. Lower concentrations were extrapolated from the calibration curve as described by Koehler et al. (5). Spiking concentrations for spiked samples (4.1.6.3) and surfaces (4.1.6.4) will be set by the gliadin protein content of the WGPAT material and according to the dilution of the spiking solution. The spiking is performed in the blank matrices prior to extraction. Incurred and contaminated samples (non-processed and processed) A naturally contaminated corn flour sample is mixed with a comparable blank sample to obtain the target values. An incurred snack sample is mixed with a comparable blank sample, respectively. Both materials were tested at five different concentration levels (including zero) with 20 repeats and one extraction per repeat (also 20 separate extractions in total). These samples were partly also used for both collaborative tests under the supervision of AACC. 4.1.6.2
