To provide a documented procedure for the determination of total anthocyanin by HPLC, with Iprona/Artemis’s methodology, expressed as cyanidin-3- glucoside. This assay can be used to determine total anthocyanin in the standardized elderberry, chokeberry and bilberry extracts. This is a reverse-phase HPLC method that can be used to calculate the total anthocyanin content of an extract as well as to examine the qualitative “fingerprint” of the anthocyanins present in the extract. This is a straight-forward dilute and shoot HPLC method. As written does not cover the identification of all Vaccinium species, Annex I, outlined in SMPR. The method can follow as written, but there are no QC/acceptability guidelines in the method. This is a simple, straight-forward method that could be readily run by most laboratories, using common reagents and instrumentation. Only one of the two main standards used in this method (Cy-glu), it should also include(Cy-rut) to be more complete in the analysis of anthocyanins. The use of two different wavelengths is not acceptable when data is being calculated from a different wavelength than the standard curve.
