The sample is extracted with acetonitrile by rapid mixing through vortexing. The liquid portion is pipetted onto a wire mesh sample support and the solvent allowed to evaporate. The mesh is mounted on the holder and it moves at a fixed speed between the dart source exit and the API entrance of the mass spectrometer. The fixed speed assures there is sufficient time to produce ions for the mass spectrometer to analyze. Data is collected in positive and negative ion mode. Qualitative assay for phosphodiesterase type 5 (PDE5) inhibitors in dietary ingredients and supplements including herbs or other botanicals; a dietary substance for use by man to supplement the diet by increasing total dietary intake; or a concentrate, metabolite, constituent, extract, or combination of any of the above dietary ingredients. {United States Federal Food Drug and Cosmetic Act §201(ff) [U.S.C. 321 (ff)]}. Dietary supplements are defined as a product intended for ingestion that contains a “dietary ingredient” intended to add further nutritional value to (supplement) the diet. Dietary supplements may be found in many forms such as tablets, capsules, softgels, gelcaps, liquids, or powders. As the supplements are predominantly natural products the potential for interference is to be minimized by optimizing the extraction solvent in order to suppress matrix interference with the assay’s ability to detect target compounds.
