Total Collaborative Study Protocol_Solus One Salmonella v1 1
aliquots used for combinations 3 and 4. Additionally, ten 26 h undiluted and diluted aliquots were used
for factorial combinations 5 and 6, with another ten 26 h undiluted and diluted aliquots used for
combinations 7 and 8.
Each combination was analyzed by the Solus One Salmonella automated preparation method, with
ten replicates of the neat and diluted inoculated aliquots and ten replicates of the undiluted non‐
inoculated aliquots, in a randomized blind coded fashion.
Results were decoded with POD values and confidence intervals calculated for combinations 1–8
when compared to the nominal combination 9. Data was analyzed for variable detection due to changes
in parameter settings.
The POD analysis of Solus One Salmonella method when tested against neat inoculated and non‐
inoculated test samples, and diluted inoculated test samples to the LOD 50
of the method, indicated that
there was no significant difference at the 5% level between nominal and experimental combinations
using the automated preparation method. A summary of Robustness POD analysis is presented in Table
6.
