All PTM and preliminary matrix evaluations were all tested using the extended confirmation. i. No, it is not acceptable. C. If the test kit enrichment is incubated at 41.5C in the collaborative study (EU temps), is it acceptable to make the claim for 42C in the US? Both have a +/- 1C range. i. Yes, it is acceptable. D. Another temperature example is for the selective agar plates and additional confirmation media required by the MLG. Can 37C (EU temp) be used and still be acceptable (as MLG states 35 +/- 1C)? I know ISPAM approved equivalence of 35 and 37 for Salmonella primary enrichments. i. 36 ± 2C E. For confirmation, is it acceptable to select one colony from one agar plate (per enrichment) to follow through to full ID? Once again, since the samples are spiked, we are most likely going to find the target organism without more than one attempt. i. Yes, as long as it is confirmed.
